Sample Prep for DNA Sequencing

Please use the following guide when preparing samples for sequencing at CUGI
  • High Copy Plasmids Bacterial Culture: Please grow an overnight culture in 96-well format in LB+15% glycerol. Seal plate with a clear plate sealer (not foil type), freeze to -80C and ship on dry ice (For on-campus customers: plate out on a petri dish so that I can pick single colonies)
  • Low Copy Plasmids Bacterial Culture: Please grow an overnight culture in 96-well format in LB+15% glycerol. Seal plate with a clear plate sealer (not foil type), freeze to -80C and ship on dry ice (For on-campus customers: plate out on a petri dish so that I can pick single colonies)
  • Purified High Copy Plasmids: Please send purified plasmid DNA at a concentration of 30-50ng either dried down or resuspended within 3µl
  • Purified Low Copy Plasmids: Please send purified plasmid DNA at a concentration of 150-200ng either dried down or resuspended within 5µl (Add 0.5ul of 50uM primer for total rxn volume of 5.5ul)
  • Purified PCR Products: Samples must be purified of unincorporated nucleotides and residual primers. RULE OF THUMB: For each kb of fragment size send approximately 10ng either dried down or resuspended within 3µl of ddH20. (Add 0.5ul of 2uM primer for total rxn volume of 3.5ul)
  • Ready to Load Samples: Send samples dried down in 96-well format. CUGI will resuspend in HiDi, denature, and load on ABI sequencer (these samples already have the Big Dye reaction completed, are cleaned up, dry and ready for directly loading on the sequencer)

On-Campus Customers
We are located 3rd Floor of the BRC. There will be an ice bucket located near the back stairwell (not the glass stairwell) for you to drop your samples off. We ask that you please drop off a copy of your order with the samples. Please follow the signs to the location – or knock on lab 302 for assistance.
