Proteomics
The Proteomics group has a focus on protein identification, characterization
and quantitation. We process biological samples from plant, animal and cell
lines. Our 2-D protein profiling uses samples for example, with characteristic
sets of proteins expressed, by specific cell types and subsets of these proteins
that are dynamically changed in response to various abiotic and biotic cues.
Gel-based and HPLC separation techniques coupled with the sensitivity and accuracy
of the mass spectrometry are used to identify these proteins and how they have
changed in regards to mass, modification (e.g. glycosylation and phosphorylation)
and quantity.
The CUGI Proteomics lab suite includes a hybrid quadrupole time of flight
mass spectrometer (Q-Tof, Waters) with electrospray ionization (ESI MS/MS) sample
introduction interfaced with a capillary liquid chromatograph (cap-LC). This
instrument has detection sensitivity for pure protein at femtomole range and
mass accuracy at 5 ppm. Additional lab equipment includes various 1-D and 2-D
gel systems and a stand-alone Waters analytical-LC with dual wavelength of
UV/RI detection systems coupled with an auto-sampler and fraction collector
in 96well format. The lab also contains a full range of supporting protein
sample preparatory equipments along with computational support through the
CUGI Bioinformatics group.
The mass spectrometry lab also provides service for small molecule analysis;
including molecular weight determination by LC-MS, purity assessment, and
quantitative analysis of synthetic and natural compounds. We provide analytical
support for method development in separation and purification of compounds of
interest from biological or synthetic material. We are experienced with photochemical,
lipids, and glycan structure and identification work.

We do peptide de novo sequencing from in-liquid and in-gel protein samples. In addition to tryptic digestion, we also perform digestion with Glu-C and Chymotrypsin routinely. The price per protein sample (gel band or pure protein in liquid) is $60 for on campus customers. Please contact us for off-campus rates. The turn around time is typically within 3 working days. The results include database search and list of identified hits.
Digest of protein with trypsin and other enzymes to generate peak list to search against known proteins in protein databases. Please contact us for rates.
We do 1D (SDS/PAGE) and 2D (Isoelectric focusing and PAGE) gel electrophoresis for protein samples in expression profiling study. The protein spots on the gel are cut, tryptic digested, and sequenced. Quantitative imaging analysis is available. The price for on campus customer is $300/two gels. We offer analysis on protein post-translational modification such as phosphorylation and glycosylation. The results include type of modification, site of occupation and degree of modification. We also do theoretical predictions using MPMAW on PTMs as well as on protein's charge, hydrophilicity, cleavage site etc. that relates to the physiochemical properties of your protein of interest. We do protein fractionation using our analytical HPLC and FPLC system using reverse phase, ion-exchange and size exclusion column chromatography in studies in the shotgun/MudPIT proteomics. The sub fractions of interests are processed with amino acid labeling techniques of your choice for subsequent quantitative analysis. Please contact us for rates.
We offer qualitative and quantitative analysis on small molecules. It includes molecular weight determination and purity assessment as well as purification from crude mixtures. We are experienced with analysis of lipids (neutral lipids, polar lipids (phospholipids), phytochemicals (plant phenolics, animal metabolites), and carbohydrates.
